近期,武漢大學(xué)黃衛(wèi)華團(tuán)隊(duì)、廈門大學(xué)Christian Amatore團(tuán)隊(duì)合作在《Nature Nanotechnology》(IF=38.8)上發(fā)表題為“Nanosensor detection of reactive oxygen and nitrogen species leakage in frustrated phagocytosis of nanofibres”(納米電極監(jiān)測(cè)巨噬細(xì)胞受挫性吞噬納米纖維過程中ROS/RNS泄漏)的研究,報(bào)告了一種電化學(xué)納米傳感器,用于監(jiān)測(cè)和定量表征單個(gè)巨噬細(xì)胞對(duì)玻璃納米纖維的吞噬作用受阻期間活性物質(zhì)釋放的通量和動(dòng)態(tài)。
機(jī)體吸入環(huán)境暴露的高長(zhǎng)徑比惰性納米纖維(石棉、玻璃纖維、碳納米管等)后,肺泡巨噬細(xì)胞會(huì)迅速識(shí)別這些異物,并試圖將其吞噬。由于纖維狀材料的高長(zhǎng)徑比和難于分解的特性,巨噬細(xì)胞難以將其快速清除,從而形成受挫性吞噬作用(frustrated phagocytosis)。在此過程中,巨噬細(xì)胞內(nèi)的NADPH氧化酶(NOX)和誘導(dǎo)型一氧化氮合酶(iNOS)會(huì)持續(xù)產(chǎn)生活性氧和活性氮(ROS/RNS)消化纖維。由于吞噬腔未封閉,大量產(chǎn)生的ROS/RNS可能會(huì)從吞噬口泄漏到胞外,對(duì)周圍肺組織細(xì)胞造成損傷并引發(fā)疾病。在過去幾十年里,研究人員采用ROS/RNS染色以及活體實(shí)驗(yàn)對(duì)碳納米管和石棉等納米纖維材料的研究均支持這一假設(shè)。然而,因缺乏合適的傳感器,該過程中各類活性物質(zhì)的化學(xué)組成及其時(shí)空動(dòng)態(tài)變化特征仍然未知。

針對(duì)受挫性吞噬作用假說,研究團(tuán)隊(duì)將鉑黑納米線電化學(xué)傳感器(SiC@PtNWS)定位在巨噬細(xì)胞吞噬口處,從而實(shí)現(xiàn)對(duì)受挫性吞玻璃納米纖維的過程中4種主要ROS/RNS(ONOO-,H2O2,NO和NO2-)泄漏通量的實(shí)時(shí)定量監(jiān)測(cè)。結(jié)果表明,在吞噬各階段,ROS/RNS釋放存在明顯時(shí)間差異。相較于細(xì)胞頂部,吞噬口處存在大量ROS/RNS泄漏,且隨吞噬進(jìn)行持續(xù)增加直到吞噬口封閉泄漏。NOX和iNOS參與動(dòng)態(tài)調(diào)控ROS/RNS的物質(zhì)組成。相關(guān)數(shù)據(jù)定量展示了單個(gè)巨噬細(xì)胞受挫性吞噬作用玻璃納米纖維時(shí)ROS/RNS產(chǎn)生的時(shí)空特征,從而描繪出巨噬細(xì)胞應(yīng)對(duì)惰性高長(zhǎng)徑納米材料的動(dòng)態(tài)免疫調(diào)節(jié)過程。體外共培養(yǎng)和活體實(shí)驗(yàn)進(jìn)一步表明,持續(xù)且大量的ROS/RNS泄漏會(huì)導(dǎo)致外周細(xì)胞嚴(yán)重受損,最終轉(zhuǎn)化為慢性炎癥和肺損傷。
