STING分子在最初發(fā)現(xiàn)時只被認為是CDS信號通路中的一個普通接頭蛋白,進一步研究表明它是環(huán)二核苷酸cyclic dinucleotide(CDNs)的直接感受分子。CDNs是細菌合成的重要第二信使,調(diào)節(jié)原核生物及哺乳動物的眾多生物進程,能啟動宿主天然免疫反應。CDNs和山酮衍生物xanthenone derivative(例如DMXAA),可結(jié)合并活化STING,通過TBK1-IRF3依賴的信號通路來誘導I型干擾素表達。
